G8P[1]型牛轮状病毒的分离和基因组特征
Isolation and genomic characterization of G8P[1]bovine rotavirus

Abstract
The purpose of this study was to isolate bovine group A rotavirus(BRVA)in yaks,a sig-nificant pathogen responsible for diarrhea in calves,and to analyze the genetic and evolutionary char-acteristics of its genome.The supernatant suspension from 3 samples of yak feces exhibiting diarrhea,which was tested positive for BRVA via RT-PCR,was filtered,and inoculated MA-104 cells.One cytopatho-genic strain was successfully isolated.This strain was identified as group A rotavirus by RT-PCR,indirect immunofluorescence,and electron microscopy.The virus titer was 105.833 TCID50/0.1 mL after plaque purification.Eleven gene segments from the genome of this strain were obtained successfully.Genotype combination of the isolate is G8-P[1]-I2-R2-C2-M2-A11-N2-T6-E2-H3,and it was named RB-8 strain.Based on the nucleotide sequences and phylogenetic trees of the 11 gene segments,RB-8 strain was a reassortment strain of group A rotaviruses,originating from bovine,human,and sheep sources.This strain may have the potential for interspecies transmission to humans.Additionally,the VP7 and VP4 proteins of the RB-8 strain exhibited unique amino acid mutations that may affect the virulence and antigenicity of this strain.This is the first G8P[1]BRVA genome from China to be deposited in GenBank,marking the initial detection of G8P[1]BRVA in yaks.This finding enhances our understanding of the prevalence and genetic evolution of BRVA.关键词
