为了研究塞内加谷病毒(SVV)VP1蛋白对细胞因子的影响,本研究通过RT-PCR、双酶切和连接试验构建表达VP1蛋白真核质粒,免疫印迹试验鉴定VP1是否正确表达,荧光定量试验检测VP1过表达对细胞因子IL-1α、IL-1β、CCL-2、CCL-5和TNF-α的影响.双酶切结果显示VP1真核质粒构建成功,遂被命名为pEGFP-C1-VP1.免疫印迹试验显示VP1蛋白能够正确表达.荧光定量试验结果显示VP1蛋白在转染后第24小时显著促进IL-1α的转录,在第12小时和第36小时分别显著促进和抑制IL-1β的转录,在第12小时和第24小时显著促进CCL-2的转录,在第12、24、36小时显著促进CCL-5的转录,在第12小时和第24小时显著促进TNF-α的转录.结果表明,SVV VP1蛋白能够诱导IL1α、CCL-2、CCL-5、TNF-α的转录;对于IL-1β,SVVVP1蛋白早期促进IL-1β的转录,后期抑制IL-1β的转录.

In order to investigate the effect of Seneca Valley virus(SVV)VP1 on cytokines,this study constructed eukaryotic plasmid expressing VP1 protein through RT-PCR,double enzyme digestion and ligation assay.Western-blot assay was used to identify whether VP1 was correctly expressed,and fluorescence quantification assay was used to detect the effects of VP1 overexpression on cytokines IL-1α,IL-1β,CCL-2,CCL-5,and TNF-α.The double enzyme digestion assay result showed that the construc-tion of VP1 eukaryotic plasmids was successful,named pEGFP-C1-VP1.Fluorescence quantification assay results showed that VP1 protein significantly promoted the transcription of IL-1α at hour 24 post-transfection,significantly promoted and inhibited the transcription of IL-1β at hour 12,24 and 36,significantly promoted the transcription of CCL-2 at hour 12 and 24,significantly promoted the transcription of CCL-5 at hour 12,24 and 36,and significantly promoted the transcription of TNF-α at hour 12 and 24.The results indicate that SVV VP1 protein can induce transcription of IL-1α,CCL-2,CCL-5,and TNF-α;For IL-1β,SVV VP1 protein promotes IL-1β transcription in the early stage and in-hibits IL-1β transcription in the later stage.
