黄芪多糖粉对湖羊瘤胃体外发酵参数和微生物区系的影响
Effects of Astragalus Polysaccharide Powder on Rumen Fermentation Parameters and Microflora of Hu Sheep in Vitro

Abstract
This experiment was conducted to investigate the effects of different adding proportions of Astraga-lus polysaccharide powder on rumen fermentation parameters and microflora of Hu sheep in vitro.Four healthy 4-month-old Hu sheep with similar body weight were selected as rumen fluid donor animals.Using a single-fac-tor experimental design,5 groups were randomly divided with 6 replicates per group and supplemented with 0(control),1.0%,1.5%,2.0%and 2.5%Astragalus polysaccharide powder in the basal diet,respectively.After 48 h in vitro fermentation,the gas and methane production,fermentation parameters,nutrient degrada-tion rates and microflora were measured,and the multiple-factor associative effects index(MFAEI)was used to evaluate the combined effects of Astragalus polysaccharide powder on rumen fermentation of Hu sheep in vitro.The results showed as follows:1)compared with the control group,the gas production in 1.5%Astraga-lus polysaccharide powder group at 12 h was significantly increased(P<0.05),the slow gas production frac-tion and potential gas production were significantly increased(P<0.05);the methane production in 1.5%and 2.5%Astragalus polysaccharide powder groups at 24 h was significantly increased(P<0.05).2)Compared with the control group,the ammonia nitrogen(NH3-N)content in fermentation broth in 1.5%Astragalus poly-saccharide powder group was significantly decreased(P<0.05),the total volatile fatty acid(TVFA)content and molar ratio of propionate in fermentation broth were significantly increased(P<0.05);at the same time,the acetate to propionate ratio in fermentation broth in 1.5%Astragalus polysaccharide powder group was the lowest,which was significantly lower than that in the other 4 groups(P<0.05).3)The crude protein degrada-tion rate in 1.5%Astragalus polysaccharide powder group was the highest,reaching 51.73%,which was sig-nificantly higher than that in the other 4 groups(P<0.05).4)The order of MFAEI of all Astragalus polysac-charide powder groups from high to low was 1.5%Astragalus polysaccharide powder group>2.0%Astragalus polysaccharide powder group>1.0%Astragalus polysaccharide powder group>2.5%Astragalus polysaccharide powder group.5)Compared with the control group,Chao1 index,observed operational taxonomic unit(OTU)number,Shannon index and Simpson index of rumen fermentation microflora in 1.5%Astragalus pol-ysaccharide powder group were significantly increased(P<0.05).At phylum level,compared with the control group,the relative abundances of Euryarchaeota,Desulfobacterota and Actinobacteriota in 1.5%Astragalus polysaccharide powder group were significantly increased(P<0.05),while the relative abundances of Spiro-chaetota,Proteobacteria and Fibrobacterota were significantly decreased(P<0.05).At genus level,compared with the control group,the relative abundances of Ruminococcus,Methanobrevibacter and Desulfovibrio in 1.5%Astragalus polysaccharide powder group were significantly increased(P<0.05),while the relative abun-dances of UCG-005 and UCG-002 were significantly decreased(P<0.05).In conclusion,dietary supplemen-tation of 1.5%Astragalus polysaccharide powder can increase the gas and methane productions in rumen fer-mentation of Hu sheep in vitro,increase crude protein degradation rate,TVFA content and propionate molar ratio,decrease NH3-N content,and improve the rumen microflora structure.[Chinese Journal of Animal Nu-trition,2025,37(3):1880-1892]关键词
