Effects of Compound Yeast Culture and β-Glucan on Growth Performance and Rumen Epithelial Barrier and Microbiota of Lambs

Abstract
This experiment was conducted to investigate the effects of compound yeast culture and β-glucan on growth performance and rumen epithelial barrier and microbiota of lambs.A total of 90 healthy 50-day-old male lambs with similar body weight were randomly divided into 3 groups with 5 replicates per group and 6 sheep per replicate.Sheep in the control group(Con group)were fed a basal diet,those in the compound yeast culture group(S1 group)were fed the diet supplemented with 50 g/kg compound yeast culture in the basal di-et,and those in the β-glucan group(S2 group)were fed the basal diet supplemented with 2 g/kg yeast β-glu-can.The pre-trial period lasted for 7 days and the experimental period lasted for 60 days.The results showed as follows:1)from days 1 to 30,compared with S2 group,the average daily gain of lambs in S1 group was sig-nificantly increased(P<0.05);from days 31 to 60,compared with Con group,the average daily gain in S1 group was significantly increased(P<0.05).2)Compared with Con group,the rumen papilla width and sur-face area of lambs in S1 and S2 groups were significantly increased(P<0.05),and the rumen papilla length in S1 group was significantly increased(P<0.05).3)Compared with Con group,the mRNA relative expression levels of claudin-1,claudin-4,occludin and zonula occludens-1(ZO-1)in rumen epithelium of lambs in S1 group were extremely significantly increased(P<0.01),the occludin mRNA relative expression level in rumen epithelium in S2 group was extremely significantly increased(P<0.01).4)Compared with Con group,the Shannon index and Simpson index of rumen microbiota of lambs in S1 and S2 groups were significantly or ex-tremely significantly decreased(P<0.05 or P<0.01).Compared with Con group,the Succinivibrionaceae_UCG-001 relative abundance in rumen in S1 group was extremely significantly increased(P<0.01),the Ag-athobacter relative abundance in rumen in S2 group was extremely significantly increased(P<0.01),and the relative abundances of Treponema,Prevotellaceae_UCG-001 and Rikenellaceae_RC9_gut_group in S1 and S2 groups were significantly or extremely significantly decreased(P<0.05 or P<0.01).5)Spearman correlation analysis showed that the claudin-1 mRNA relative expression level in rumen epithelium was significantly posi-tively correlated with the Succinivibrionaceae_UCG-001 relative abundance in rumen(P<0.05),and signifi-cantly negatively correlated with the relative abundances of Prevotellaceae_UCG-001,Treponema and Rikenel-laceae_RC9_gut_group(P<0.05);the mRNA relative expression levels of occludin and ZO-1 in rumen epi-thelium were extremely significantly negatively correlated with the Prevotellaceae_UCG-001 relative abundance in rumen(P<0.01).In conclusion,dietary compound yeast culture can increase the average daily gain of lambs,promote the growth and development of rumen epithelium and the tight junction protein expression,and regulate the tight junction protein expression in rumen epithelium by regulating the relative abundances of spe-cific bacteria in rumen,thus enhancing the rumen epithelial barrier function,which is better than β-glucan.[Chinese Journal of Animal Nutrition,2025,37(3):1904-1918]关键词
