Research progress in E3 ubiquitin ligase FBXW2 and its clinical relevant diseases

Abstract
FBXW2,a member of the F-box and WD repeat domaining(FBXW)protein family,is an important component of the Skp1-Cullin1-F-box(SCF)E3 ubiquitin ligase complex.In recent years,its molecular characteristics and biological functions have attracted increasing attention.As an E3 ubiquitin ligase,FBXW2 participates in the regulation of various protein levels,playing crucial roles in cell cycle progression,maintenance of genomic stability,and determination of cell fate,It is closely associated with the development and treatment of numerous diseases.For instance,FBXW2 can function as a tumor suppressor,inhibiting the progression of various cancers such as lung cancer,breast cancer,and prostate cancer.Additionally,FBXW2 acts as an inflammatory mediator in macrophages and is involved in the formation of atherosclerosis and insulin resistance-related obesity.FBXW2 is also implicated in periosteal osteogenesis.This review provides a detailed summary of the physiological mechanisms of FBXW2 and its pathophysiological roles in diseases such as lung cancer,breast cancer,and atherosclerosis,aiming to offer a foundation for future research of FBXW2.关键词
