Abstract
PDZ domain-containing protein 1(PDZK1)is a scaffold protein with multiple PDZ domains that plays a critical role in regulating transmembrane transport proteins,ion channels,and signal transduction proteins.It is ultimately involved in lipid metabolism,cell proliferation,differentiation,migration,tumor growth,and ion transport.In renal cell carcinoma(RCC),particularly in clear cell renal cell carcinoma(ccRCC),PDZK1 expression is significantly downregulated and is associated with tumor progression and poor prognosis.Upregulating PDZK1 expression not only suppresses tumor cell growth and slows disease progression,but also modulates the phosphorylation pathway of platelet-derived growth factor receptor beta(PDGFR-β),thereby improving drug resistance.This may benefit patients with advanced or metastatic RCC who rely primarily on molecular targeted therapies.Moreover,PDZK1 holds promise as a biomarker for personalized treatment of RCC and as a novel therapeutic target,with potential in predicting treatment response.关键词
