Preliminary evaluation of the biological function of non-coding RNA BprsB in Burkholderia pseudomallei

Abstract
Objective:To investigate the effects of Hfq related sRNA(BprsB)on the growth rate,motility,biofilm forma-tion,and drug resistance of Burkholderia pseudomallei.Method:Using homologous recombination technology,BprsB of Burk-holderia pseudomallei dependent on Hfq protein was used as the target to construct a BprsB mutantt strain(ΔBprsB);The wild-type Burkholderia pseudomallei strain(HNBP001)was used as the control group,and the ΔBprsB was used as the experi-mental group;The bacterial growth curve was measured by a multimode reader to evaluate changes in growth rate;Using point plate culture method and measuring the diameter of the colony to evaluate motility;The formation of biofilm was measured by crystal violet staining method;Finally,the sensitivity of bacteria to antibiotics is determined through MIC experiments.Result:The growth rate of ΔBprsB was lower than that of HNBP001;Compared with HNBP001,the population diameter of ΔBprsB was smaller(P<0.000 1);The biofilm formation of ΔBprsB was less than that of HNBP001(P<0.000 1);Compared with HN-BP001,the ΔBprsB was more sensitive to quinolone antibiotics(Levofloxacin,Norfloxacin,Ciprofloxacin)and β-lactam antibiot-ics(Aztreonam).Conclusion:sRNA BprsB promotes the growth rate,motility,biofilm formation,and drug resistance of B.pseu-domallei,laying a solid foundation for further research on the role of sRNA BprsB in the pathogenesis of B.pseudomallei.关键词
