Abstract

Canine herpesvirus(CHV)has a high infection and mortality rate in newborn puppies,causing serious economic losses to the dog breeding industry.Currently,there are few reports on this virus,and there is a lack of effective vaccines for prevention and control of the virus infection.A strain of the virus was isolated from sample collected from a sick dog using Madin-Darby Canine Kidney(MDCK)cells,which named as CHV-202402.The isolated strain was identified through methods such as observation of viral particle morphology,indirect immunofluorescence(IFA)assay,Western blot,and homology analysis of gene sequences.The results showed that the CHV-202402 strain could produce typical CPE on MDCK cells,with a viral titer of 10-4 TCID50/0.1 mL at the third passage;spherical,enveloped viral particles could be observed under the electron microscope;IFA and Western blot results indicated that the isolated strain could specifically bind with the monoclonal antibodies against CHV;homology analysis of the gD gene nucleotide sequence showed that it was highly homologous to that of domestic and international strains.These results proved that the isolated strain was the canine herpesvirus.The successful solation of a CHV strain has enriched the genomic data of CHV and laid the foundation for studying the characteristics of CHV and establishing related detection methods.
