抗犬细小病毒多克隆抗体的制备与鉴定
Preparation and Characterization of Polyclonal Antibodies against Canine Parvovirus

Abstract
To explore the effectiveness of polyclonal antibodies(pAb)in detecting and identifying various species of parvovirus,canine parvovirus CPV-2 was cultured in F81 cells.After virus purification using sucrose density gradient centrifugation,polyclonal antibodies were generated by immunizing New Zealand white rabbits.The antibody serum was collected,and the antibodies were purified through ammonium sulfate saturation and Protein A affinity chromatography to obtain rabbit polyclonal antibodies against CPV-2.The ELISA titer of the CPV-2 rabbit pAb was 1∶160 000,and the hemagglutination inhibition titer was 1∶217.Immunofluorescence assays showed that the CPV-2 rabbit pAb could recognize different species of parvovirus,including canine parvovirus,feline panleukopenia virus,and ferret enteritis virus,but did not react with other canine pathogens,indicating broad-spectrum detection ability and specificity of the pAb.Western blot test analysis demonstrated that the CPV-2 rabbit pAb could detect the structural proteins VP1,VP2,VP3,and the non-structural proteins NS1 and NS2 of canine parvovirus.Virus neutralization tests in vitro showed that the CPV-2 rabbit pAb had neutralizing activity against canine parvovirus,with a neutralizing titer of 1∶26.364 at a concentration of 1 mg/mL,which suggested that the CPV-2 rabbit pAb had potential therapeutic values.The prepared CPV-2 rabbit pAb with good reactivity,strong specificity,and broad detection spectrum,provides a foundation for enhancing parvovirus detection,diagnosis,and therapeutic formulations.关键词
