The Application of CAR-T Cell Therapy in Autoimmune Diseases
摘要
Abstract
CAR-T cell therapy,an innovative immunotherapy,has achieved remarkable advancements in hematological malig-nancies.It has also shown great potential and broad application in the field of autoimmune diseases.CD19 is a common marker of B cells.CAR-T cells targeting CD19 can eliminate abnormal B cells and autoantibodies,achieving a"reset"of the immune system.It has achieved great effectiveness in the treatment of diseases such as systemic lupus erythematosus(SLE).Patients can achieve complete remission of clinical symptoms,serum conversion of auto-antibodies,and relapse-free status even after discontinuation of immunosup-pressive drugs.BMCA CAR-T cells targeting plasma cells have also shown great interest in recent years.Emerging CAAR-T cell and CATCR-T cell therapies bring more possibilities for precision treatment.In clinical application,it is crucial to select the appropriate disease types,patients,and targets,and closely monitor various parameters after CAR-T cell infusion.Although currently still facing challenges such as cytokine release storm and immune-mediated neurotoxicity,immunosuppression,and applicability in multi-organ involvement diseases,through exploration and optimization of various strategies,it is expected to gradually overcome these shortcom-ings.The future research of universal allogeneic CAR-T cells and in-depth research in the scientific fields,such as CAR structure opti-mization,target screening,side effect control,and cost-effectiveness evaluation will continuously promote the development of CAR-T cell therapy,ultimately change the therapeutic landscape of autoimmune diseases.关键词
