GLP-1受体激动剂对动脉粥样硬化小鼠血脂、炎性因子和氧化应激水平的影响
Effect of GLP-1 Receptor Agonists on Blood Lipid Levels,Inflammatory Factors,and Oxidative Stress in ApoE-/-Mice

Abstract
Objective To investigate the effects of glucagon-like peptide-1(GLP-1)receptor agonists on the levels of lipids,inflammatory factors,and oxidative stress in ApoE knockout(ApoE-/-)atherosclerosis(AS)mice.Methods ApoE-/-mice were randomly divided into three groups:a control group,a low-dose group,and a high-dose group.There were 10 mice in each group.The mice were fed with western diet for 8 weeks before injection of liraglutide.As a control group,ten C57BL/6J mice were given a regular diet for 8 weeks before being injected with normal saline.The hematoxylin-eosin(HE)staining technique was applied to assess the thickness of the intima-media(IMT)and fibrous cap in atherosclerotic(AS)lesions.Enzyme-linked immunosorbent assay(ELISA)was utilized to detect expression levels of TC,TG,LDL-C,HDL-C,TNF-α,IL-6,and IL-1β.Xanthine oxidase and thio-barbital(TBA)methods were employed to detect SOD and MDA levels in mice.Results Compared with the control group,the IMT and plaque fibrous cap thickness increased(P<0.05),and levels of LDL-C,TG,TC,IL-6,IL-1β,and TNF-α levels in serum increased(P<0.05),while HDL-C levels decreased,MDA increased,and SOD decreased in the model group(P<0.05).Compared with the model group,the IMT levels decreased(P<0.05),the fibrous cap thickness increased(P<0.05),the serum LDL-C,TG,TC,IL-6,IL-1β,and TNF-α levels decreased(P<0.05),the HDL-C levels increased(P<0.05),the GSH decreased(P<0.05)both in the low and high dose groups.Compared with the model group,there was no statistically significant decrease in MDA or increase in SOD in the low-dose group(P>0.05),but the decrease in MDA and increase in SOD in the high-dose group were more obvious(P<0.05).Compared with the low-dose group,the high-dose group showed no significant changes in aortic morphology,blood lipid levels,inflammatory factor levels,or oxidative stress levels(P>0.05).Conclusion Liraglutide may enhance the stability of atherosclerotic plaques,potentially through its impact on improving blood lipid profiles,modulating inflammatory responses,and exerting antioxidant effects in mice.关键词
