末水条斑紫菜对健康C57BL/6J小鼠肠道微生物的影响
Effect of last stage Porphyra yezoensis on intestinal microbiota of healthy C57BL/6J mice

Abstract
[Objectives]The aim of this study was to investigate the effects of last stage Porphyra haitanensis on the intestinal microbiota and their metabolic products in healthy C57BL/6J mice.[Methods]The mice were gavaged with saline(control group),1.15 g·kg-1(low-dose group),and 2.30 g·kg-1(high-dose group)doses of last stage P.yezoensis.After 28 days,mouse samples were collected for pathological section observation,short-chain fatty acid content determination,and intestinal microbiota 16S rDNA high-throughput sequencing.[Results]The results indicated no significant changes in the growth performance of mice gavaged with last stage P.haitanensis.Compared to the control group with a colon length of 53.50 mm,the colon length in the low-dose group significantly increased to 65.60 mm.Pathological sections of the colon tissue revealed that last stage P.haitanensis increased the number of goblet cells.The results of short-chain fatty acids in the colonic contents showed no significant changes in the levels of acetic acid,propionic acid,n-valeric acid,and isovaleric acid,while the level of butyric acid of low-dose group significantly increased,and the levels of butyric acid and isobutyric acid of high-dose group significantly increased.The result of 16S rDNA in the colonic contents found that both low-dose and high-dose administration of last stage P.yezoensis reduced the diversity of intestinal microbial diversity,increased the relative abundance of Firmicutes and decreased that of Bacteroidetes.However,the relative proportion of the genus Dubosiella significantly increased.Pathogens such as Norank_f_norank_o_Clostridia_UCG-014 and Helicobacter showed a decreased relative abundance.[Conclusions]Last stage P.haitanensis could regulate the intestinal microbiota of healthy C57BL/6J mice,promoting the growth of beneficial bacteria and the production of butyric acid.The optimal intake was 1.15 g·kg-1.关键词
