基于网络药理学探讨逍遥散改善SSRIs抵抗机制
Exploring Mechanisms of Action of Xiaoyao San on Improving Resistance to SSRIs Based on Network Pharmacology

Abstract
Based on the network pharmacology,this study explored the efficacy,characteristics,and mechanism of Xiaoyao San in improving the resistance of SSRIs in treatment of depression.Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP),Online Mendelian Inheritance in Man(OMIM),GeneCards,and UniProt databases were applied to ob-tain the active ingredients of Xiaoyao San and the targets to improve the resistance of five SSRIs including Fluvoxamine,Fluoxetine,Paroxetine,Sertraline,and Citalopram on depression.STRING website was used to obtain the protein interaction network.Metascape website was used to enrich the potential targets.A visual network diagram of"Xiaoyao San-potential target-pathway-SS-RIs resistance"was created by using Cytoscape software.The results demonstrated that the core active ingredients of Xiaoyao San were peony glycoside,β-sitosterol,kaempferol,luteol,etc.PPI network showed that BCL2,ESR1,and NOS2 were the common core targets for Xiaoyao San to improve the resistance of SSRIs.These core targets mainly acted on AGE-RAGE signaling pathway in diabetic complications,chemical carcinogenesis-receptor activation,Apelin signaling pathway and HIF-1 signaling pathway,regu-lating cellular response to organic cyclic compound,negative regulation of cell population proliferation,response to inorganic sub-stance,which played a role in improving the resistance effects of SSRIs on depression.The present results confirmed that Xiaoyao San improved the resistance effects of five SSRIs on depression through multiple components on multiple targets and pathways.Si-multaneously,they provided a theoretical basis for the rational and effective clinical combination of drugs.关键词
