Exploring the Mechanism of Secondary Components of Huangqi Injection in the Treatment of Leukopenia Using Network Pharmacology

摘要

Abstract

The objective of this study was to investigate the active components in secondary components of Huangqi injection and its mechanism for the effect on leukopenia using network pharmacology,molecular docking and in vivo experiments.According to the mass spectrometry information,the cleavage law of the reference substance and the reference literatures,the chemical compositions in secondary components of Huangqi injection were identified;Swiss Target Prediction platform was used to obtain the relevant tar-gets of the druggable components;leukopenia-related targets was screened by Gene cards and Disgenet databases;the intersections between the obtained component targets and disease-related targets were used to obtain the potential targets of secondary compo-nents of Huangqi injection in treating leukopenia,and the"component targets-disease targets"network was constructed by Cyto-scape 3.9.1 software to analyze the active ingredients;the common targets were imported into the String website for protein-protein interaction analysis to identify key targets;Gene Ontology(GO)function and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway were carried out through bioinformatics website;molecular docking and leukopenia mouse models were used to verify the results of network pharmacology.A total of 25 compounds were identified in secondary components of Huangqi injection,including 21 flavonoids and 4 saponins.There are 121 potential targets for secondary components of Huangqi injection in treating leukopenia,involving 486 biological processes,68 cellular components,96 molecular functions and 155 signaling pathways.Five potential ac-tive ingredients including Isorhamnetin-3-O-β-gentiobioside,Astragalin,6"-O-acetylCalycosin-7-O-D-glycoside,6"-O-acetylono-nin,and Wogonin,8 key target proteins including TP53,AKT1,IL6 and signaling pathways such as Phosphatidylinositol-3-kinase/protein kinase B(PI3K-Akt)signaling pathway,Janus kinase-signal transducer and activator of transcription(JAK-STAT)signaling pathway were obtained.The 5 potential active ingredients have strong binding affinity to the key targets.Secondary components of Huangqi injection could significantly regulate the contents of IL-6 and G-CSF in serum of leukopenia mice.Isorhamnetin-3-O-β-gentiobioside,Astragalin,6"-O-acetylCalycosin-7-O-D-glycoside,6"-O-acetylononin,and Wogonin are the active components of secondary components of Huangqi injection,which may act on key targets such as TP53,AKT1,IL6,etc,key pathways including PI3K-Akt signaling pathway,and JAK-STAT signaling pathway to play its therapeutic leukopenia.
