柴胡桂枝汤抗抑郁的网络药理与孟德尔随机化研究
Network Pharmacology and Mendelian Randomization Study on the Antidepressant Effects of Chaihu Guizhi Decoction

Abstract
This study aims to comprehensively utilize network pharmacology,Mendelian randomization analysis,and molecular docking technology to deeply explore the specific mechanism of Chaihu Guizhi Docoction(CZT)in treating depression.Firstly,we systematically collected the drug targets of CZT and the disease targets of depression,and determined the common key targets through intersection target gene analysis and protein-protein interaction screening.Subsequently,Gene Ontology(GO)functional analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis were employed to deeply analyze the biological pathways involved in these targets.To initially validate the screened core targets,molecular docking technology was utilized.Additionally,Mendelian randomization methods from the OpenGWAS database were used to further verify and deepen the screening results.The results showed that we identified 193 major active components of CZT and screened out 30 potential targets,which may be closely related to the effect of CZT in treating depression.The application of molecular docking technology further confirmed that active components such as Formononetin,Isorhamnetin,Quercetin,Stigmasterol,and Wogonin have good affinity with targets such as TNF,AKT1,IL6,IL 1 β,and PTGS.In the Mendelian randomization analysis,we found 10 and 17 single nucleo-tide polymorphisms(SNPs)associated with depression in the genome-wide association analysis data for AKT1 and IL 1β targets,re-spectively(both with P<5×10-5).After strict screening,we retained 7 and 15 SNPs for the final Mendelian randomization analysis.The analysis results indicated that AKT1 and IL 1β are likely to be key targets for depression treatment.This study revealed that CZT plays an important role in treating depression by acting on targets such as TNF,AKT1,IL6,IL 1β,and PTGS.This discovery pro-vides a solid theoretical foundation and research direction for the clinical application of CZT in the field of depression treatment.关键词
