基于转录组学联合蛋白质组学的加味七方胃痛颗粒抗慢性萎缩性胃炎的作用机制探究
Exploring the action mechanism of modified Qifang Weitong Granule against chronic atrophic gastritis based on transcriptomics combined with proteomics

Abstract
Objective To investigate the action mechanism of modified Qifang Weitong Granule(QFWTG)on chronic atrophic gastritis(CAG)using transcriptomics combined with proteomics.Methods Histopathological and morphological changes in the gastric tissues of rats from each group were observed by hematoxylin-eosin(HE)staining.Differentially expressed genes(DEGs)and differentially ex-pressed proteins(DEPs)were screened using transcriptomics and quantitative proteomics techniques.Then,co-omics and enrichment analyses were performed,followed by verification of the signaling pathways and key proteins identified through protein immunoblotting.Results Gastric mucosal atrophy was significantly relieved in the positive control and herbal groups.Omics testing revealed 479 shared DEGs and 37 shared DEPs in the comparisons between the blank group and the model group,as well as between the herbal group and the model group.The integrative omics analysis of the model and herbal groups identified nine proteins/genes with mapped relationships,pri-marily involved in the mitogen-activated protein kinase(MAPK)signaling pathway.Experimental results demonstrated that modified QFWTG reduced the expression levels of HSPA4L,ERK,JNK,and RAS in the rat gastric tissues,consistent with the findings from the integrative omics analysis.Conclusion Modified QFWTG can reverse the gastric mucosal atrophy in CAG model rats.The mechanism is related to the inhibited protein expressions of HSPA4L,ERK,JNK,and RAS and the regulation of the MAPK signaling pathway.关键词
