低氧条件下丹栀逍遥散对乳腺癌细胞增殖侵袭及缺氧相关因子的影响
Exploring the Effect of Danzhi Xiaoyao Powder on proliferation and invasion and hypoxia-re-lated factors of breast cancer cells under hypoxic conditions

Abstract
Objective To investigate the effects of Danzhi Xiaoyao Powder(DZXYP)-containing serum on the proliferation,invasion and expression of hypoxia-related factors in MDA-MB-231 breast cancer cells under hypoxic conditions.Methods Cell proliferation was assessed using the MTT assay.MDA-MB-231 cells were divided into the normoxic control group,the hypoxic control group,and the DZXYP group(with serum oncentrations of 10%,15%,and 20%).The cells were cultured for 24,48,and 72 hours to observe the inhibitory effects of the drug on cell proliferation.A scratch assay was performed to assess the cell migration and invasion in each group.Western blot(WB)was used to detect the protein expression levels of hypoxia-inducible factor-1α(HIF-1α),monocarboxylate transporters 1/4(MCT1/4),caveolin-1(CAV-1),and CXC chemokine receptor 4(CXCR4)in each group.Results The MTT as-say results indicated that under hypoxic conditions,cell proliferation was significantly inhibited in the DZXYP group(P<0.05),with the highest inhibition rate observed at 48 hours using 20% serum.The scratch assay results showed a significant decrease in cell migra-tion rate in the DZXYP group(20%)(P<0.01).WB analysis revealed that under hypoxic conditions,the protein expression levels of HIF-1α,MCT1,MCT4,and CXCR4 were decreased in the DZXYP group(20%)(P<0.05,P<0.01),while the expression of CAV-1 was increased(P<0.05).Conclusion DZXYP-containing serum can significantly inhibit the proliferation,migration and in-vasion of MDA-MB-231 breast cancer cells under hypoxic conditions,thus exerting anti-cancer effects.This may be related to its regulation of hypoxia-associated lactate metabolism and epithelial-mesenchymal transition(EMT).关键词
