Abstract

Objective To investigate the influence of interleukin-33(IL-33)on the transcriptional level in rat cardiac microvascular endothelial cells(CMECs).Methods Rat CMECs were isolated and cultured,then divided into a control group and an IL-33 group(add IL-33 with a final concentration of 50 ng/mL).Transcriptome sequencing was conducted to analyze the differential gene expression and related pathway enrichment between the two groups.Quantitative PCR was employed to confirm the expression of relevant genes.Finally,a co-expression network analysis of differential expressed genes was used to explore the regulatory network of rat CMECs induced by IL-33.Results After IL-33 induction of CMECs,a total of 103 upregulated genes and 97 downregulated genes were identified(P<0.05).The expression levels of the chemokine ligand 2(Ccl2)and chemokine ligand 1(Cxcl1)genes were upregulated(P<0.05),while the expression levels of vascular cell adhesion molecule 1(Vcam1)and intercelluar adhesion molecule-1(Icam1)showed no significant changes(P>0.05).Functional clustering analysis and pathway enrichment analysis of differentially expressed genes revealed that IL-33 upregulated genes associated with granulocyte chemotaxis and migration,while downregulated genes related to cardiomyocyte action potentials.Conclusion IL-33 regulates a substantial number of genes associated with the function of rat CMECs,suggesting its role in the cardiac microenvironment.
