Abstract

To study the anti-depressive effects of different doses of purslane alcohol extract on mice,and to preliminarily explore its anti-depressive and alleviating animal stress response mechanisms.The effective substances of purslane were extracted using an ultrasound-assisted extractor.Mice were randomly divided into groups and a reserpine-induced mouse depression model was established.Each dose group of mice was orally administered with purslane alcohol extract(high dose group 600 mg/kg,medium dose group 300 mg/kg,low dose group 150 mg/kg)for 14 consecutive days.The anti-depressive effect was evaluated by behavioral observation,and the changes in 5-HT,NE,and DA levels in the hippocampus of mice were detected by ELISA to preliminarily explore the anti-depressive mechanism of purslane.In the reserpine-induced depression model,the low,medium,and high dose groups of purslane alcohol extract could significantly reduce the impact of reserpine on the body temperature of mice(P<0.05),and also reduce the eyelid drooping score.The high dose group of purslane alcohol extract showed significant difference compared to the model group(P<0.05).Additionally,all groups could significantly increase the content of 5-HT,NE,and DA in the hippocampus of mice.Purslane alcohol extract has anti-depressive pharmacological activity,which may be related to enhancing the function of the 5-HT,NE,and DA systems.
