乳黄消对LTA和无乳链球菌作用牛乳腺上皮细胞病理模型的抑制效果研究
Anti-inflammatory Intervention Effect Study of Ruhuangxiao on the Infection of Bovine Mammary Epithelial Cells by LTA and Streptococcus Agalactiae

Abstract
In order to explore the mechanism of Ruhuangxiao in the treatment of mastitis in dairy cows,the inhibitory effect of Ruhuangxiao on the pathological model of bovine mammary epithelial cells(BMECs)treated with LTA and Streptococcus agalactiae(S.agalactiae)was carried out.The expression levels of inflammatory factors in BMECs treated with LTA at concentrations of 15µg/mL and 20µg/mL for 12 h and S.agalactiae MOI of 25,50,100 for 0 h,2 h and 4 h were detected by qPCR to establish the best modeling conditions.On this basis,the experiment was divided into two experimental groups of Ruhuangxiao+S.agalactiae and Ruhuangxiao+LTA,as well as three control groups of LTA,S.agalactiae and blank.The optimal concentration of Ruhuangxiao was 0.312 5 mg/mL for pretreatment of BMECs for 12 h and 24 h by qPCR,and then BMECs were treated with 15µg/mL LTA and S.agalactiae with MOI of 50 for 12 h and 2 h,respectively,to explore the inhibitory effect of Ruhuangxiao on LTA and S.agalactiae on BMECs.The results of the inhibitory effect of Ruhuangxiao on inflammatory cells showed that 0.312 5 mg/mL Ruhuangxiao could significantly down-regulate the expression of TNF-α,IL-8,IL-1β and IL-6 inflammatory factors caused by LTA and S.agalactiae,while the inflammatory factors in the two control groups without Ruhuangxiao intervention had no downward trend.This study shows that Ruhuangxiao can significantly reduce the inflammatory damage of BMECs caused by LTA and S.agalactiae,thereby alleviating the inflammatory response.关键词
