Cloning and Tissue Expression Analysis of TMEM182 Gene in Yak

Abstract
To elucidate the biological structure and function of the TMEM182 gene in yaks,as well as its expression profile across various tissues,this study utilized cDNA derived from the muscle tissue of domestic yaks to clone the CDS region of the TMEM182 gene.Subsequently,a comprehensive bioinformatics assessment was carried out.Finally,the expression levels of the TMEM182 gene were determined in lung,kidney,heart,liver,muscle,spleen,testis,and adi-pose tissue of yak.The results revealed that The CDS region sequence length of the TMEM182 gene in domestic yak is 690 bp,encoding 299 amino acids.Nucleotide sequence alignment indicated that homology between domestic yak and wild yak was the highest at 99.7%,while it was the lowest with macaques at 85.9%.Bioinformatics analysis addition-ally revealed that the theoretical isoelectric point of the TMEM182 protein in domestic yak is 6.48;it is characterized as a slightly acidic hydrophobic protein lacking a signal peptide but possessing transmembrane domains,and mainly dis-tributed in the endoplasmic reticulum.This protein has 12 potential phosphorylation sites and 4 glycosylation sites.The secondary structure mainly comprises random coils.TMEM182 protein interacts significantly with proteins such as TM6SF1,MCTP2,TSPAN33,and CDH15.Results from qRT-PCR indicated varying degrees of expression for the TMEM182 gene across all examined tissues in domestic yak,in which notably higher expression levels were observed in testis,muscle,and adipose tissues compared to other tissues(P<0.01).关键词
