低温处理下日本医蛭及其环带组织转录组差异表达基因分析OA
Analysis of differentially expressed genes in transcriptome of Hirudo nippnica whitman and its cliellum tissue under low temperature treatment
[目的]探究低温对日本医蛭(Hirudo nippnica)不同组织中相关基因表达的影响.[方法]将 40 尾日本医蛭随机均分为 2组,分别置于常温 24℃和低温 6℃恒温培养箱内饥饿处理 28 d后,采集环带、肌肉和头部进行转录组测序,分析不同温度下 3 种组织中差异表达基因并进行筛选及信号通路富集,同时采用荧光定量 PCR方法对差异基因进行验证.[结果]不同温度下,环带、肌肉和头部中各有差异表达基因 2 465个、2 588个和 1 998个,其中 737个差异表达基因为 3个组织共有.GO分析显示生物过程中色氨酸代谢途径以及分子功能中相关酶活性的基因显著富集.KEGG分析表明低温影响了多条与能量代谢相关的通路(PPAR、AMPK、NF-kappa、BGnRH)和不饱和脂肪酸的转化相关差异基因的表达.此外,环带中LDLR、CaM、notch、PLK1和ADIPOR等繁殖相关基因表达上调.[结论]低温处理使日本医蛭代谢和内分泌通路相关基因大量表达,这些基因参与了色氨酸的代谢、脂肪酸的转化以及生殖细胞的增殖,为解析低温介导日本医蛭繁殖机理提供数据支撑.
[Objective]This study aimed to investigate the effects of low temperature on the expression of related genes in different tissues of Hirudo nippnica.[Method]Forty H.nipponica were randomly divided into two groups,three tissues(cliellum,muscle and head)of H.nippnica were sequenced,assembled and annotated at normal temperature(24℃)and low temperature(6℃)conditions with starvation for 28 days,and the dif-ferentially expressed genes(DEGs)of different tissues were analyzed by GO and KEGG databases,meanwhile,the DEGs were validated by qPCR.[Result]Under different temperatures,there were 2 465,2 588 and 1 998 DEGs in cliellum,muscle and head,respectively,of which 737 DEGs were shared by three tissues.Gene on-tology analysis indicates that the gene expression of related tryptophan metabolism and enzyme activity in bio-logical process and molecular function were significant.KEGG pathway analysis suggests that hypothermia affected many pathways related to energy metabolism(PPAR,AMPK,NF-kappa,BGnRH)and the conversion of unsaturated fatty acids,as well as the expression of related DEGs.LDLR,CaM,notch,Plk1 and ADIPOR genes were up-regulated in the cliellum,which may be involved in the development and regulation of the gonad of H.nippnica.[Conclusion]These differential genes are involved in the metabolism of tryptophan,the trans-formation of fatty acids and the proliferation of germ cells under low temperature treatment,this study underpin understanding the mechanism of low temperature mediated reproduction of H.nippnica.
