应用大蒜素缓解牛子宫内膜炎损伤的研究
Study on Alleviating Bovine Endometritis Injury with Allicin
摘要
Abstract
[Objective]To study the treatment effect of allicin on bovine endometritis injury.[Method]Bovine endometrial epithelial cells were induced by lipopolysaccharide to establish an inflammatory cell model,and explore the anti-inflammatory mechanism of allicin.The opti-mal concentration of LPS was screened out.The secretion,mRNA relative expression and protein relative expression of inflammatory factors were detected by ELISA,fluorescence quantitative PCR and Western blot method.[Result]Different concentrations of LPS could improve cell proliferation,and the cell viability was the highest when LPS concentration was 30 μg/mL.Compared with inflammatory cells,allicin could in-hibit the secretion of inflammatory factors TNF-α,IL-1β,IL-6 and IL-8 in LPS-induced endometrial epithelial cells,and also reduced the gene transcription levels of IL-1β,TLR4,and NF-κβ p65.Allicin inhibited the expression of TLR4 and NF-κβ p65 proteins by Western blot a-nalysis.[Conclusion]Allicin can inhibit the TLR4/NF-κβ signaling pathway and exert anti-inflammatory activity by regulating the secretion of inflammatory cytokines in cells.关键词
