番茄bHLH转录因子SlbHLH077的克隆及表达分析

Abstract
As one of the largest gene families in eukaryotes,bHLH transcription factors play a key role in plant growth and development and abiotic stress.In order to further study the function and expression characteristics of the bHLH transcription factor in tomato,this study used to-mato seedlings as the test material,cloned the SlbHLH077 gene with a CDS full length of 1 739 bp in tomato,and the basic characteristics of this gene were analyzed by bioinformatics methods.Combined with real-time fluorescence quantitative technology to study the gene expression pattern under biotic and abiotic stress.The results show that the gene contains only one exon,no introns,and encodes 579 amino acids,con-taining a typical HLH conserved domain.The relative molecular weight of the encoded protein is 64 271.00 Da,and the theoretical isoelectric point is 7.59,which is an unstable basic protein.Protein interaction indicates that the protein interacts with 10 JAZ proteins to play a mediating role.The phylogeny indicated that SlbHLH077 has the closest relationship with potato and the farthest relationship with rhododendron.Tissue a-nalysis showed that the gene was expressed in multiple tissues,and fluorescence quantitative PCR showed that the gene responded to abiotic stresses such as low temperature,salt and drought,and biological stresses such as hormones and yellow leaf curl disease.The results of this study provide a theoretical basis for tomato resistance breeding and further exploration of the function of SlbHLH077.关键词
