高效液相色谱法测定膨化宠物食品中姜黄素类化合物的含量
Determination of curcumin compounds in puffed pet food by high-performance liquid chromatography

Abstract
An efficient high-performance liquid chromatography(HPLC)method was established for the determination of curcumin,demethoxycurcumin,and bisdemethoxycurcumin in dog feed.The HPLC was performed using a C18 column(250 mm×4.6 mm×5 μm)with a mobile phase consisting of acetonitrile and 0.1%formic acid in a ratio of 65:35.Methanol was used as the extraction solvent,with a flow rate of 1.0 mL/min,a column temperature of 30℃,and a detection wavelength of 425 nm.A sample volume of 10 μL was injected.The curcumin compounds were dissolved in methanol using ultrasonic extraction for 30 minutes,followed by fil-tration before analysis.The results indicated that the method exhibited good linearity for curcumin,demethoxycurcumin,and bis-demethoxycurcumin in the concentration ranges of 4.24~84.83 μg/mL,1.70~68.00 μg/mL,and 5.64~84.56 μg/mL,respectively.The limits of detection were 0.020 μg/mL,0.013 μg/mL,and 0.017 μg/mL,while the quantification limits were 0.080 μg/mL,0.0544 μg/mL,and 0.068 μg/mL,respectively.This method is easy-operation,cost-efficient,highly accurate,and with excellent precision,which is proper for the determination of curcumin compounds in dog feed.关键词
