基于LC-MS/MS技术测定类风湿性关节炎患者血浆中微量氨甲蝶呤浓度方法学的建立
Development of an assay for measurement of serum trace methotrexate in patients with rheumatoid arthritis by LC-MS/MS

Abstract
Objective:To develop a liquid chromatography-tandem mass spectrometry(LC-MS/MS)method capable for measuring serum methotrexate in RA patients.Methods:With methotrexate-d3,the chromatography was performed on Agilent EC-C18(2.1 mm×50 mm,2.7 μm)column with mobile phase consisting of 0.1%formic acid-water and acetonitrile.The flow rate was 0.3 mL·min-1.Column temperature:40℃.Results:The retention time of methotrexate was 2.939 min.The linear range of methotrexate plasma samples was 0.001 2～3 μmol·L-1(r2=0.999 9).Conclusion:The LC-MS/MS method established in this study is accurate,simple and rapid.It could provide basis for clinical individualized medication.关键词
