Causal relationship between dairy intake and sarcopenia risk:A two-sample Mendelian randomization study

Abstract
Objective:Observational studies suggest that dairy intake may have a protective effect against sarcopenia,but the causal relationship between dairy consumption and the risk of sarcopenia remains unclear.This study aims to evaluate the causal relationship between dairy intake and sarcopenia using a two-sample Mendelian randomization(MR)analysis,to provide reference for clinical prevention and treatment. Methods:Summary-level data for dairy consumption(cheese,milk,yogurt,and butter)and sarcopenia-related phenotypes(appendicular lean mass,low handgrip strength,and walking pace)were obtained from the IEU open Genome-Wide Association Studies(GWAS)database.Two-sample MR analysis was performed to assess the causal relationship,with inverse variance-weighted(IVW)as the primary method and MR-Egger,weighted median(WME),simple mode(SM),and weighted mode(WM)as complementary methods. Results:IVW analysis showed causal associations between dairy intake and reduced risk of sarcopenia:cheese intake was positively associated with walking pace(OR=1.185,95%CI 1.153 to 1.219,P<0.001)and negatively associated with low handgrip strength(OR=0.757,95%CI 0.658 to 0.871,P<0.001);butter intake was positively associated with walking pace(OR=1.179,95%CI 1.007 to 1.380,P=0.041).Sensitivity analyses supported the robustness of these causal relationships.However,no causal associations were found between milk,yogurt,or butter intake and appendicular lean mass(P=0.059,P=0.768,P=0.661,respectively),or low handgrip strength(P=0.550,P=0.986,P=0.288,respectively),nor between milk/yogurt intake and walking pace(P=0.895,P=0.365,respectively). Conclusion:Cheese intake is causally associated with increased handgrip strength and faster walking pace,and butter intake is causally associated with faster walking pace.There are no causal relationships between other types of dairy products and reduced risk of sarcopenia.关键词
