猪圆环病毒2型灭活疫苗的植物源免疫增强剂筛选OA北大核心

Screening of plant-based immune enhancers for inactivated porcine circovirus type 2 vaccine

[目的]筛选安全、高效的抗猪圆环病毒2型病毒疫苗免疫增强剂,改善现有佐剂的免疫效力及安全性.[方法]以昆明小鼠为研究对象,选取萜类(青蒿素、紫杉醇、穿心莲内酯、熊果酸)、生物碱类(黄连素、石蒜碱、千金藤素、氧化苦参碱)、酚酸类(咖啡酸、绿原酸、没食子酸、香豆酸)和黄酮类(山柰酚、槲皮素、木犀草素、原花青素)共16种植物源活性物质,通过测定小鼠体质量比例、IgG抗体水平、脏器指数、总超氧化物歧化酶(total super-oxide dismutase,T-SOD)、丙二醛(malondialdehyde,MDA)和总抗氧化能力(total antioxidant capacity,T-AOC),开展靶向猪圆环病毒2型灭活疫苗免疫增强剂的筛选,并对其抗氧化活性进行初步评价.[结果]1)小鼠灌胃植物源活性物质后,体质量呈增加趋势.2)各组抗体水平呈现先上升后下降的趋势.与疫苗组相比,黄连素、石蒜碱、千金藤素、槲皮素、木犀草素和原花青素组可极显著性提高机体的抗体水平,其中以原花青素组效果最佳,比疫苗组抗体水平提高了59.59%.3)槲皮素组小鼠心脏、肝脏和肾脏指数呈显著性升高,黄连素、石蒜碱、原花青素组在统计学意义上无显著差异;其中,槲皮素、原花青素组小鼠脾脏和胸腺指数呈极显著性提高,脾脏指数提高了49.50%和53.23%,胸腺指数提高了121.82%和103.64%,提示其可能在促进免疫器官的发育、分化和成熟,提高机体的免疫功能方面发挥重要作用.4)木犀草素、槲皮素和原花青素组T-SOD活性呈极显著性升高,MDA含量呈极显著性降低,千金藤素、石蒜碱、槲皮素、木犀草素、黄连素和原花青素组T-AOC呈极显著性升高.以原花青素对机体的抗氧化效果较好,T-SOD活性和T-AOC分别提高了 9.60%和 48.57%,MDA含量降低了26.82%.[结论]原花青素对小鼠的免疫增强效果最佳,可作为靶向猪圆环病毒2型灭活疫苗佐剂研究的候选者.

[Objective]The aim of this study was to screen a safe and efficient immune booster against porcine circovirus type 2 vaccine,and to improve the immune efficacy and safety of existing adjuvants.[Method]In this study,Kunming mice were selected as the research object,and a total of 16 plant-based active substances were selected:terpenoids(artemisinin,taxol,andrographolide,ursolic acid),alkaloids(berberine,lycorine,cepharanthine,oxymatrine),phenolic acids(caffeic acid,chlorogenic acid,gallic acid,coumaric acid)and flavonoids(kaempferol,quercetin,luteolin,proanthocyanidin).By measuring the body mass ratio,IgG antibody level,organ index,total superoxide dismutase(T-SOD),malondialdehyde(MDA)and total antioxidant capacity(T-AOC)of mice,the immune enhance-ment agents targeting porcine circovirus type 2 inactivated vaccine were screened and the antioxidant activity was evaluated.[Result]1)The body mass of mice increased after the administration of plant derived active substances.2)The antibody levels of all groups showed a trend of first increasing and then decreasing after vaccination.Compared with vaccine group,berberine,lycorine,cepharanthine,quercetin,luteolin and proanthocyanidin groups could significantly improve the antibody level of the body,and the proanthocyanidin group had the best effect,among which the antibody level was increased by 53.59%.3)The results of organ index displayded that the heart,liver and kidney indexes in querce-tin group were significantly increased,but there were no significant differences in berberine,lycorine and proanthocyanidin groups.The spleen and thymus indexes of quercetin and proanthocyanidin groups were significantly increased by 49.50%and 53.23%,121.82%and 103.64%,suggesting that querce-tin may play an important role in promoting the development,differentiation and maturation of immune organs and improving the immune function of the body.4)The T-SOD activity in luteolin,quercetin and proanthocyanidin groups was significantly increased,MDA content was significantly decreased,and T-AOC content was significantly increased in cepharanthine,lycorine,quercetin,luteolin,berberine and proanthocyanidin groups.The antioxidant effect of proanthocyanidins was better,T-SOD activity and T-AOC were increased by 9.60%and 48.57%,respectively,and MDA was decreased by 26.82%.[Con-clusion]Proanthocyanidins have the best immune enhancement effect on mice,and could be used as a candidate for the study of porcine circovirus type 2 inactivated vaccine adjuvant.
