基于半制备液相色谱法从槐米水提液中制备芦丁
Preparation of Rutin from Extract of Huaimi(Sophorae Flos Immaturus)in Water Based on Semi-Preparative Liquid Chromatography

Abstract
Objective:To establish a rapid preparation method of rutin from Huaimi(Sophorae Flos Immaturus)based on semi-preparative liquid chromatography,and to provide reference for the rapid preparation of rutin standard.Methods:The yield of water extract precipitates of Huaimi was taken as the index to optimize the best extraction and resting method of Huaimi water extract was optimized.Rutin was separated by semi-preparative liquid chromatography,and its purity was verified and calculated by using thin layer chromatography(TLC)and high performance liquid chromatography(HPLC).Results:The best preparation method for water extract precipitates of Huaimi was to take 2 g of Huaimi,add 15 times the amount of water,heat and reflux for 1 h,filter while hot,let the filtrate stand at 25 ℃ for 24 h,then filter and dry.Rutin was prepared by using the optimized semi-preparative liquid chromatography and verified by TLC and HPLC.The purity of rutin was calculated to be 98.62％.Conclusions:Rutin can be quickly sepa-rated from Huaimi by water extraction and semi-prepared liquid chromatography with high purity,which provides the new method and idea for-the separation and preparation of rutin.关键词
