犬疱疹病毒gD蛋白单克隆抗体的制备及抗原表位鉴定
Preparation of Monoclonal Antibodies against Canine Herpesvirus gD Protein and Identification of the Epitope

Abstract
To obtain the gD protein of canine herpesvirus(CHV)and monoclonal antibodies(mAbs)against the gD protein,and to analyze the linear antigenic epitopes recognized by these antibodies,the CHV gD gene was cloned into the pET-32a vector and transformed into BL21(DE3)competent cells.After purification,an approximately 41.5 kDa recombinant gD protein was obtained,which was consistent with the expectation.Mice were immunized with the recombinant protein,and monoclonal antibodies were prepared using cell fusion technology.Three stable hybridoma cell lines(gD1,gD2,gD3)that secreted antibodies against the gD protein were obtained.Subclass identification showed that their heavy chains were IgG2b,and light chains were κ.The gD3 mAb recognized an antigenic epitope with the amino acid sequence of 245FKEALRYDIPY255.The results enrich the antigenic epitope information of CHV and provide a foundation for the study of the function of the CHV gD protein and the development of detection methods for CHV.关键词
