Abstract

In order to understand the epidemiological characteristics and genetic evolution of feline calicivirus(FCV),samples of conjunctival and nasal swabs from cats suspected upper respiratory tract disease in a pet hospital in Changchun were collected for virus isolation.The viruses were identified using dual-Nano PCR,colloidal gold test strip,and indirect immunofluorescence method.The isolated strains were then subjected to whole-genome amplification and genetic evolution analysis.Subsequently,artificial infection experiments were conducted.The results indicated that the F81 cells in the test group exhibited the cytopathic effect(CPE)such as cell rounding,grape-like clusters,and detachment.In the immunofluorescence assay,the F81 cells in the test group displayed specific green fluorescence,indicating the successful isolation of a feline calicivirus strain,which was hereby designated as the CC19-01 strain.The homology of the CC19-01 strain with known FCVs worldwide ranged from 76.4%to 83.6%.The genome of this strain is highly variable.It had a close phylogenetic relationship with the classic strain MZ712017.1,MZ712021.1,MZ712018.1 isolated in Nanning and was on a parallel evolutionary branch with the vaccine strains JX519210.1(America)and KM111171.1(America).The results of the artificial infection experiments showed that cats developed symptoms 3 to 7 days after infection with this strain,with increased body temperature,increased ocular discharge,loss of eyelashes around the eyes,decreased appetite,and lethargy.The clinical symptoms of infected cats began to recover after 7 days.These results indicated that the genome of the CC19-01 strain of feline calicivirus was highly variable and not lethal to cats.The experiment not only enriched the basic data of domestic FCV but also provided significant molecular information for the development of FCV vaccine and the prevention and control of the disease.
