温通活血乳膏对2型糖尿病周围神经病变大鼠核因子-E2相关因子2的影响
Wentong Huoxue cream for type 2 diabetic peripheral neuropathy effect of Nrf2 in rats

Abstract
Objective:Wentong Huoxue cream was used to intervene diabetic peripheral neuropathy(DPN)rat model to observe its effect on nuclear factor E2-related factor 2(Nrf2)oxidative stress pathway.Methods:Among 36 adult Wistar male rats,6 rats were used as blank group and 30 rats were used as DPN model.The 30 rats were ran-domly divided into model group,matrix group,metformin group,medium and high dose groups of Wentong Huoxue cream.No drug intervention was given to blank group and model group,matrix group was given matrix intervention of Wentong Huoxue cream,metformin group was given metformin intervention,Wentong Huoxue cream medium and high dose groups were given medium and high dose Wentong Huoxue cream,respectively.After the last dose,serum levels of myeloperoxidase(MPO)and tumor necrosis faction-α(TNF-α)were detected by enzyme-linked immu-nosorbent assay(ELISA).Expressions of Nrf2 and NF-κB protein in sciatic ganglia were detected by Western blot.And expressions of Nrf2 and NF-κB mRNA were detected by RT-PCR.Results:Wentong Huoxue cream could de-crease the expression of MPO,TNF-α,NF-κB protein and mRNA,and increase the expression of Nrf2 protein and mRNA(all P<0.05),and the improvement effect of high dose Wentong Huoxue cream was more significant.Conclusion:Wentong Huoxue cream can inhibit the expression of NF-κB,improve the oxidative stress and play a role in the treatment of DPN by up-regulating the expression of Nrf2.关键词
