NAFLD promotes the development of psoriasis vulgaris:a study based on bidirectional Mendelian randomization

Abstract
Objective To explore the potential bidirectional causal relationship between non-alcoholic fatty liv-er disease and psoriasis vulgaris using a two-sample Mendelian randomization(MR)approach.Methods Sin-gle nucleotide polymorphism(SNP)information of the two samples was obtained from publicly available genome-wide association analysis databases,and suitable SNPs screened as instrumental variables.The causal relationship between the two samples was analyzed by two-way MR analysis using inverse variance weight-ing,Egger regression,weighted median and weighted mode,supplemented by various sensitivity analyses to calibrate the results.Results Forward MR analysis revealed that the inverse variance weighting method and the weighted median method showed a significant causal effect of nonalcoholic fatty liver disease on psoriasis vulgaris(both P<0.05),whereas the results of reverse MR did not support reverse causality.The MR-pleiotro-py function and MR-PRESSO test revealed no horizontal pleiotropy(P>0.05),Cochran's Q test revealed no heterogeneity(P>0.05),and the leave-one-out sensitivity analysis proved the stability of the MR findings.Conclusion MR analysis revealed a positive causal relationship between non-alcoholic fatty liver disease and psoriasis vulgaris.关键词
