摘要

Abstract

Objective:To investigate the effect of edaravone on melanogenesis in B16 cells induced by 3-isobutyl-1-methylxanthine(IBMX).Methods:B16 cells were treated with 75 μmol/L IBMX and then exposed to edaravone at concentrations of 0,25,and 100 μmol/L,respectively.After co-culture for 72 hours,the expression of the melanin marker HMB-45 was observed by immunohistochemistry,melanin content was measured by NaOH lysis method,tyrosinase protein expression was analyzed by Western blot,and tyrosinase activity was detected by the dopa oxidation method.Results:Under IBMX induction,the activity of tyrosinase increased,the expression of HMB-45 was upregulated,and melanin content increased in B16 cells(all P<0.05).After the addition of edaravone,tyrosinase activity decreased,the expression of HMB-45 was downregulated,and melanin content reduced,and showed a dose-dependent inhibitory effect(all P<0.05).Conclusion:Edaravone inhibits IBMX-induced melanogenesis in B16 cells,and its mechanism of action may be related to the regulation of tyrosinase activity and HMB-45 expression.
