Abstract
To investigate the pharmacokinetics of ceftiofur hydrochloride in adult healthy beagle dogs after subcutaneous ceftiofur hydrochlo-ride injection.Six adult healthy beagle dogs,half male and half female,were injected subcutaneously with 10%ceftiofur hydrochloride injec-tion at a single dose of 10 mg/kg.Then,blood samples were collected from the veins in the forelimbs of the dogs before and after administra-tion at different time points.Next,plasma samples were treated with dithioerythritol(DTE)to separate ceftiofur and its desfuroylceftiofur(DFC)metabolites from proteins or sulfur-containing compounds.After extraction and purification,the plasma concentration was determined by high performance liquid chromatography(HPLC)and quantified using the external standard method.Finally,the measured plasma con-centration was calculated by a non-compartment model.The results were as follows:The peak time(Tmax)was(7.33±3.27)h,the peak concentration(Cmax)was(33.87±7.58)μg/mL,the area under the drug time curve(AUC0-t)was(720.54±150.39)h·μg/mL,the area under the drug time curve(AUC0-∞)was(726.67±148.90)h·μg/mL,and the eliminating half-life(t1/2λz)was(16.61±5.51)h.The effective plasma concentration could be maintained for more than 4 days after a single dose.These results indicated that the method of subcutaneous injection of ceftiofur hydrochloride developed here was eliminated slowly in the body of dogs,and it could maintain the effective plasma concentration for a long time,with a sustained-release and long-acting effect.关键词
