摘要

Abstract

Chicken-derived Salmonella not only causes infections and diseases in poultry but also poses threats to food safety and public health.In this study,a strain of Salmonella,named SM24,was isolated from the tissues of a suspected Salmonella-infected chicken in An-hui Province,China.The growth curve of this strain was evaluated,its pathogenicity and median lethal dose(LD50)were determined using a specific pathogen-free(SPF)chick inoculation assay,and its resistance profile was tested usingthe Kirby-Bauer disk diffusion method.Fi-nally,whole-genome sequencing was performed to construct a genomic map of the isolate,and the characteristics of virulence and antimicro-bial resistance genes it carried were analyzed.The results showed that the isolate grew well on the LB medium,and displayed pathogenicity to SPF chicks with an LD50 of 8.50×105 CFU in the chicks.The isolate SM24 was multidrug-resistant,to penicillins,cephalosporins,quinolo-nes,tetracyclines,sulfonamides,neomycin,gentamicin,azithromycin,and aztreonam,and so on,a total of 16 antibiotics,and it was sen-sitive tospectinomycin,amikacin,chloramphenicol,florfenicol,imipenem,and polymyxin B.The whole-genome sequencing analysis re-vealed that SM24 was a Salmonella Indiana(S.Indiana)serotype,sequence type ST17,with a genome size of 4 865 511 bp and a GC con-tent of 51.96%.It harbored one IncHI2 plasmid of 165 044 bp.A total of 280 virulence-associated genes were carried by the SM24 strain;144 antimicrobial resistance genes were annotated in its genome,with 12 resistance genes located on integrative and mobilizable elements(IME)and plasmid.Furthermore,SM24 was genetically distant from the chicken-derived S.Indiana reported in China.In summary,the present isolated chicken-derived S.Indiana strain exhibited high virulence and multidrug resistance.Its whole-genome profiling facilita-ted a deeper understanding of the pathogenicity and antimicrobial resistance mechanisms of S.Indiana,providing a reference for further research.
