外周血circRNA在乙肝相关性肝硬化患者中的差异表达与诊断价值研究
Research on the differential expression and diagnostic value of circRNA in peripheral blood in patients with hepatitis B virus-related cirrhosis

Abstract
Objective To investigate the differential expression of circular RNA(circRNA)in peripheral blood in hepatitis B virus(HBV)-related cirrhosis,and screen suitable diagnostic markers to provide research basis for early diagnosis of patients with hepatitis B cirrhosis.Methods Patients diagnosed with hepatitis B and admitted to Dongguan Eighth People's Hospital,who were diagnosed with liver cirrhosis in the early stage by liver puncture and had the same liver fiber measurement value from April 2023 to April 2024 were selected and divided into the experimental group(n=10),and other healthy people in the same period were selected and divided into the control group(n=10).circRNA-seq technology was used to detect the expression profile of circRNA in the peripheral blood of the two groups,the database was used to analyze the differences between the two groups,and the appropriate differential expression of circRNA was screened as a diagnostic marker.Based on this,30 patients with hepatitis B cirrhosis and 30 healthy people were selected.The differential expression level of corresponding circRNA in peripheral blood of the two groups was verified by real-time quantitative polymerase chain reaction(RT-qPCR),and the receiver operating characteristic curve(ROC)was established to evaluate the diagnostic value of circRNA detection in peripheral blood for hepatitis B cirrhosis.Results A total of 7419 circRNAs were detected in the peripheral blood of the experimental group and the control group.Compared with the control group,349 of them in the experimental group expressed circRNA differentially,227 were up-regulated and 122 were down-regulated.hsa_circ_0005632 and hsa_circ_0101617,as two upregulated circRNA,were obviously higher in peripheral blood expression level than those in the control group(P<0.05).ROC analysis of the above two circRNA showed that the area under the curve(AUC)was 0.90 and 0.94.Both of them had good diagnostic efficiency,and hsa_circ_0101617 had superior diagnostic efficiency.Conclusion circRNA is differentially expressed in peripheral blood of patients with HBV-related cirrhosis,in which hsa_circ_0005632 and hsa_circ_0101617 can be used as new diagnostic markers of cirrhosis.关键词
