黄精调节氧化应激水平保护衰老大鼠脾脏的实验研究
Experimental Study on the Spleen Protection Effect of Polygonatum Sibiricum in Aging Rats through Regulating Oxidative Stress

Abstract
[Objective]To observe the effects of Polygonatum sibiricum(PS)on spleen index,histopathological changes of spleen tissue,oxidative stress indicators,and the expression of senescence proteins p53 and p21 in aging rats.[Methods]Ninety-six 18-month-old rats were randomly divided into an aging group and high,medium,and low-dose PS groups based on their body weight,with 24 rats in each group.Additionally,24 2-month-old rats were designated as the young group.The young and aging groups were gavaged with an equal volume of distilled water,while the high,medium,and low-dose PS groups were gavaged with 4 g/kg,2 g/kg,and 1 g/kg of PS decoction,respectively,for 12 weeks.At the 4th,8th,and 12th weeks,8 rats from each group were weighed and then executed by anesthesia.The body weight change rate and spleen index were calculated.Histopathological sections of the spleen were observed,and the levels of superoxide dismutase(SOD),malondialdehyde(MDA),and total antioxidant capacity(T-AOC),as well as the expression of p53 and p21 proteins,were detected.[Results]With increasing age,the aging group ex-hibited a decreased spleen index,reduced levels of T-AOC and SOD in spleen tissue,increased MDA levels,upregu-lated expression of p53 and p21 proteins(P<0.05 or P<0.01),and less distinct spleen tissue structure with vague boundaries between red and white pulp.Compared with the aging group,PS intervention in aging rats resulted in an in-creased spleen index,elevated levels of T-AOC and SOD in spleen tissue,decreased MDA levels,downregulated ex-pression of p53 and p21 proteins(P<0.01),and improved histopathological changes in spleen tissue.[Conclusion]Po-lygonatum sibiricum can protect the spleen tissue of aging rats by regulating oxidative stress levels.关键词
