槲皮素通过SLC7A11/GPX4/ACSL4通路调控铁死亡改善卵巢早衰的机制研究
Study on the mechanism of quercetin regulating iron death through SLC7A11/GPX4/ACSL4 pathway to improve premature ovarian failure
摘要
Abstract
Objective:To evaluate the protective effect of quercetin on premature ovarian failure(POF)and the regulatory mechanism of iron death based on POF mouse model.Methods:During July and August,2024,a total of 30 five-week-old female SPF C57BL/6 mice were randomly assigned to the normal,model,and quercetin group.Besides the normal group,the POF model was established by intraperitoneal injection of cyclophosphamide in the other two groups.Quercetin group was given querce-tin gavage for 21 d,and the other two groups were given the same dose of normal saline.After the intervention,mouse serum was taken for AMH,FSH,E2,T-SOD,and CAT determination,and ovarian tissue was taken for H&E staining,immunohistochem-ical analysis and qRT-PCR detection.Results:Compared to the normal group,the atretic follicles in the model group were signifi-cantly increased,AMH and E2 levels were significantly decreased(P<0.01),FSH levels were significantly increased(P<0.01),T-SOD activity and CAT activity were decreased significantly(P<0.01),SLC7A11 and GPX4 expressions were signifi-cantly decreased(P<0.05),and the expression of ACSL4 was significantly increased(P<0.01).Compared to the model group,atretic follicles were reduced in the quercetin group,mature follicles were observed in ovarian pathological tissue,AMH and E2 levels were significantly increased(P<0.01),and FSH level decreased significantly(P<0.01).Compared to the model group,T-SOD activity and CAT activity in quercetin group were significantly increased(P<0.05);SLC7A11 expression was in-creased,GPX4 expression was increased,and ACSL4 expression was decreased(P<0.05).Conclusion:Quercetin can inhibit iron death by activating SLC7A11/GPX4/ACSL4 pathway,thereby improving ovarian function and sex hormone level in POF mice.关键词
