天然黄酮类化合物干预DKD中NLRP3靶点研究进展
Research progress on natural flavonoids targeting the NLRP3 in DKD
摘要
Abstract
Diabetes kidney disease(DKD)is one of the progressive microvascular complications of the diabetes,which is the main cause of end-stage renal disease.More and more evidence suggest that renal inflammation plays a crucial role in the pathogen-esis of DKD,and its mechanism is related to the excessive activation of inflammatory mediators.NLR family pyrin domain contain-ing protein 3(NLRP3)inflammasome,as a classic inflammasome,can assemble into NLRP3 multimeric complexes in cells.When the body receives internal/external pathogens or injury,the NLRP3 multimeric complexes activates a series of inflammatory responses in the body,promoting the release of inflammatory factors IL-1β and IL-18,thereby triggering the occurrence and devel-opment of DKD.This article mainly emphasizes that the role of NLRP3 inflammasome in NLRP3/caspase-1/IL-1β pathway in-ducing abnormalities of renal intrinsic cells(such as tubular epithelial cells,podocytes,and mesangial cells)and the related mecha-nisms in the pathophysiology of DKD.In addition,the potential application of natural flavonoids as candidate drugs is used to selec-tively target the NLRP3 inflammasome and signaling pathway molecules for reducing the inflammatory reaction and alleviating the symptoms of DKD is discussed.The article can provide new idea for the scientific management and treatment of DKD patients.关键词
