肉苁蓉总苷拟雌激素作用的网络药理学研究
Network pharmacology study on estrogenic-like effect of glycosides of Cistanchedeserticola

Abstract
Explored the potential mechanism of estrogen-like effects of glycosides of cistanchedeserticola based on network pharmacology combined with transcriptomic data.40 active ingredients of glycosides of Cistanchedeserticola(GCs)with estrogenic effect were predicted by TCMSP,SymMap,ETCM,Stitch and QSAR databases.The target information of estrogenic deficiency disease was retrieved by Genecards and OMIM databases.Screening key targets and constructing PPI network;GO functional enrichment analysis and KEGG pathway enrichment analysis were performed to identify key targets and pathways.Combined with the preliminary transcriptomic data,the correlation analysis between network pharmacological prediction targets and KEGG pathway was conducted to further reveal the mechanism of the estrogen-like effect of GCs.A total of 183 targets were screened,and a total of 1 712 targets were selected for estrogen deficiency diseases.24 core key targets were obtained.GO function and KEGG pathway enrichment show that GCs mainly act on biological processes such as cytokine receptor binding,estrogen receptor binding,growth factor receptor binding,nuclear receptor activity,nuclear hormone receptor binding,steroid hormone receptor activity and hormone receptor binding.It regulates tumor necrosis factor(TNF)signaling pathway,interleukin-17(IL-17)signaling pathway,hypoxia-inducible factor-1(HIF-1)signaling pathway,estrogen signaling pathway and other pathways.Compared with transcriptome data,gene expression changes were detected in all 24 core targets except Mcl1 and Gcg,among which Ar,Casp8,Esr1,Fos and Hmox1 were differentially expressed genes.There are 68 KEGG pathways.Estrogen signaling pathway,cAMP signaling pathway,TNF signaling pathway was the main signaling pathway.Glycosides of Cistanchedeserticola can exert estrogen-like effects through"multi-component,multi-target,multi-pathway",and the mechanism may be related to the regulation of estrogen signaling pathway,cAMP signaling pathway and TNF signaling pathway.关键词
