Effects of Heat Treatment of Embryonated Eggs on Growth,Immunity and Microbiota in Chicks

Abstract
To investigate the effects of heat treatment of embryonated eggs on the growth,immunity and microbiota of layer chicks,an experiment was conducted using fertile White Leghorn eggs.The eggs were randomly divided into control and heat treatment(HT)groups with at least 3 replicates per group and 25 eggs per replicate.The heat treatment lasted for 6 days during the hatching process.The results showed that heat treatment of embryonated eggs could significantly decrease hatchability,the higher treatment temperature,the lower hatching rate,but the embryonic periods of heat treatment had a lesser effect.Besides,compared with the control group,the relative chick weight and brain,liver,lung weight were all significantly decreased in the HT group,while yolk weight was significantly increased.Further analysis of the RNA-seq results indicated that 583,1 775 and 871 differentially expressed genes respectively in hypothalamus,liver and intestine were enriched for the GO terms cytokine pathway,inflammatory response,immune defense and so on.In addition,heat treatment of embryonated eggs reduced the survival rate of chicks when injected with LPS.151,2 259 and 589 genes were respectively induced in hypothalamus,liver and intestine under LPS response.Among them,73,1 370 and 129 genes significantly affected by heat treatment were enriched in the immune related pathways.Moreover,compared with the control group,the bacteria in intestine tract and yolk fluid of chicks in the HT group were significantly increased,and intestinal lactobacillus and oscillospira were decreased,enterococcus was increased.In summary,heat treatment of embryonated eggs could reduce hatchability,retard growth,weaken immunity and change intestinal microbiota structure in chicks.关键词
