黄芪甲苷通过上调miR-1226缓解ox-LDL诱导人脐静脉血管内皮细胞损伤
Effects of Astragaloside Ⅳ on ox-LDL-Induced Endothelial Cell Injury in Human Umbilical Vein by Upregulating miR-1226

Abstract
Objective:To discuss the effect of astragaloside Ⅳ on injury of human umbilical vein endothelial cells(HUVECs)and its mechanism.Methods:HUVECs were cultured in vitro and treated with 100 mg/L ox-LDL to establish atherosclerosis(As)cell mod-el.HUVECs were divided into blank control group,model group,low,medium and high doses astragaloside Ⅳ groups,miR-NC group,miR-1226 group,high dose astragaloside Ⅳ with anti-miR-NC group,high dose astragaloside Ⅳ with anti-miR-1226 group.Flow cytom-etry was used to detect apoptosis.ELISA method was used to detected IL-6,IL-1β,TNF-α levels;The protein expression was detected by Western Blot.The expression of miR-1226 was detected by RT-qPCR.Results:Compared with blank control group,apoptosis rate,Bax protein expression and the secretion of IL-6,IL-1β and TNF-α were significantly increased in model group,while expressions of Bcl-2 protein and miR-1226 were significantly decreased(P<0.05).Compared with the model group,the apoptosis rate,Bax protein expres-sion and the secretion of IL-6,IL-1β and TNF-α in each astragaloside Ⅳ concentration group and miR-1226 group were significantly in-creased,the expression of Bcl-2 protein was significantly decreased,and the expressions of Bcl-2 protein and miR-1226 were significant-ly increased(P<0.05).The effect of astragaloside Ⅳ was concentration-dependent.Down-regulation of miR-1226 reversed the effect of astragaloside Ⅳ.Conclusion:Astragaloside Ⅳ may alleviate ox-LOL-induced HUVECs injury by up-regulating the expression of miR-1226.关键词
