国产5种杠柳属植物的分子鉴定与系统发育分析
Molecular Identification and Phylogenetic Analysis of Five Domestic Plants of Periploca

Abstract
Objective:To identify five species of Periploca by DNA barcoding,to clarify the phylogenetic relationship of Periploca sepium,Periploca forrestii,Periploca tsangii,Periploca calophylla,Periploca floribunda.Methods:The DNA genome of five plants of Periploca was extracted by modified CTAB method,and PE150 bp double-terminal sequencing was performed by Illumina HiSeq X-Ten sequencing platform.The Maximum-Likelihood(ML)method was used to construct phylogenetic trees for ITS2 sequence and chloroplast genome sequence,respectively.Results:The ITS2 sequence length of five plants of Periploca was 267 bp,and the GC%was 55.7%to 56.6%.Phylogenetic trees analysis showed that five plants of Periploca were clustered into monophyletic clades,among which Periploca forrestii and Periploca tsangii were clustered into one branch,Periploca sepium was clustered into Periploca floribunda,and Periploca calophylla was clustered into another branch.The length of chloroplast genes ranged from 153 513 to 154 140 bp,GC%was 38.1%to 38.2%,the total number of genes was 128 to 132.Phylogenetic tree analysis showed that the five plants of Periploca were monophyletic clades,among them,a monophyletic branch was formed by Periploca floribunda and Periploca tsangii,and the other three species were monophyletic clades.Conclusion:ML trees based on ITS2 sequence can distinguish Periploca plants from other genera,and the support rate of differentiation among intra-genus plants is low.The ML tree based on chloroplast genome can distinguish the plants in the genus Periploca.Therefore,the chloroplast genome is more suitable for the identification of Periploca species than ITS2 sequence.关键词
