摘要

Abstract

Objective:To study the genetic diversity of ITS segment in 6 species of Pedicularis,and to analyze the feasibility of using this segment as DNA barcode to identify Pedicularis.Methods:Total DNA of Pedicularis was extracted,and ITS sequence of nucle-ar genome was amplified by PCR and sequenced.Chromas 1.0 software was used to collate the sequence and determine ITS sequence.Genetic distances of 20 closely related plants were calculated using MEGA 11.0,phylogenetic trees of adjacency(NJ)and maximum parsimony(MP)were constructed and compared.Results:The ITS sequences of 6 species of Pedicularis were amplified and sequenced successfully.The ITS sequence information of Pedicularis rhinanthoides subsp.labellata was submitted to the NCBI database,and the GenBank entry number(OQ102382)was obtained.The average genetic distance of 20 species of Pedicularis was 0.1109.Compared with NJ method,the phylogenetic tree constructed by MP method was more consistent to the results of traditional methed of classification of Pedicularis.Conclusion:ITS sequences can be used as DNA barcode to distinguish and identify 6 species of Pedicularis.
