Comparative Study on Fingerprint and Chroma Value of Mori Cortex Containing Raw Bark and Peeling Raw Bark

Abstract
Objective:To compare the difference of Mori Cortex containing raw bark and peeling raw bark.Methods:A total of 12 batches of Mori Cortex from different habitats were collected,the fingerprints of Mori Cortex containing raw bark and peeling raw bark were established by UPLC,the similarity evaluation was carried out.The chroma values(L*,a*,b*)of each batch of Mori Cortex pow-der were determined,and the average total chroma values E* was calculated.The one-way ANOVA,principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA)were used to perform multivariate statistical analysis.Re-sults:The established fingerprints of Mori Cortex containing raw bark and peeling raw bark all had 23 common peaks,and chromato-graphic peaks 6,13,18 were identified as mulberroside A,oxyresveratrol,kuwanon G,respectively.The similarity between the finger-prints of 12 batches of Mori Cortex containing raw bark and peeling raw bark was all greater than 0.900.The results of one-way ANOVA showed that the chromatographic peaks 1,3,15 to 23 and the colorimetric values L*,a*,E* were significantly different between the fingerprints of Mori Cortex containing raw bark and peeling raw bark(P<0.05).The chromatographic peaks 2,4,7,11,14 were posi-tively correlated with colorimetric values a*,b*,while the chromatographic peak 7 was negatively correlated with L*.PCA could be di-vided into two kinds of most Mori Cortex containing raw bark and peeling raw bark.The comprehensive score of principal components of most Mori Cortex containing raw bark was higher than that of Mori Cortex peeling raw bark.According to the VIP>1,the peaks 1,3,15 to 23 were the main difference components between Mori Cortex containing raw bark and peeling raw bark.Conclusion:The established UPLC fingerprint method of Mori Cortex is stable and feasible,and there are differences in the fingerprints and chroma values of Mori Cortex containing raw bark and peeling raw bark.关键词
