摘要

Abstract

Objective:To investigate the effect and mechanism of atractonolide Ⅲ on H2O2-induced inflammatory response of H9c2 cardiomyocytes.Methods:H9c2 cells were induced by H2O2 to establish inflammatory response model.The experiment was divided into blank control group,model group and atractonolide Ⅲ(15,30,60 μmol/L)groups.Mitochondrial ROS level was detected by cellular immunofluorescence.The mRNA expressions of IL-1β,TNF-α,iNOS,IL-4,IL-10 and Arg-1 were detected by qPCR.The expressions of IL-1β,TNF-α,iNOS,TRIM38,NAP1,IRF3 were detected by Western Blot.TRIM38 expression was down-regulated by transfection with siRNA,the effects of atractonolide Ⅲ on mitochondrial ROS,IL-1β,TNF-α,iNOS mRNA and NAP1,IRF3 protein expressions in H9c2 cells under H2O2 were observed.Results:Compared with blank control group,mitochondrial ROS production in model group was signifi-cantly increased,IL-1β,TNF-α,iNOS mRNA and IL-1β,TNF-α,iNOS,NAP1,IRF3 protein expressions were significantly increased.The expressions of IL-4,IL-10,Arg-1 mRNA and TRIM38 protein were significantly decreased(P<0.01).Compared with the model group,the intervention of atractonolide Ⅲ could significantly reduce the generation of ROS in mitochondria,and significantly decrease the expressions of IL-1β,TNF-α,iNOS mRNA and IL-1β,TNF-α,iNOS,NAP1,IRF3 proteins.The expressions of IL-4,IL-10,Arg-1 mRNA and TRIM38 protein were significantly increased(P<0.05 or P<0.01).After TRIM38 was down-regulated,the above effects of atractonolide Ⅲ were weakened.Conclusion:Atractonolide Ⅲ regulates the balance between TRIM38-NAP of cardiomyocytes,inhibits downstream IRF3 activation,reduces mitochondrial ROS release,promotes anti-inflammatory signaling,inhibits inflammation.
