Abstract
Objective:To explore the mechanism of Dan'e Fukang Decoction in the treatment of endometriosis(EMS)based on network pharmacol-ogy and validate the findings through animal experiments.Methods:The TCMSP database was used to screen for the effective components and corresponding targets of Dan'e Fukang Decoction.Disease targets were obtained by querying the GeneCards,OMIM,and other databases using"Endometriosis"as a keyword.A"disease-active ingredient-target"network was constructed by taking the intersection of drug and disease targets.The mechanism of Dan'e Fukang Decoction in the treatment of EMS was predicted through bioinformatics analysis.An animal model of EMS was constructed to evaluate and validate the therapeutic effect and mechanism of Dan'e Fukang Decoction.Results:Network pharma-cology screening identified 47 major active ingredients in Dan'e Fukang Decoction and 107 drug-disease cross-targets.Key targets included signal transducer and activator of transcription 3(STAT3),nuclear factor kappa-B(NF-κB),mitogen-activated protein kinase(MAPK),and hypoxia-inducible factor 1α(HIF-1α).GO enrichment analysis mainly focused on epithelial cell proliferation,regulation of transcription fac-tors,and cytokine receptor activity.KEGG enrichment analysis mainly involved the advanced glycation end products and their receptor(AGE-RAGE)signaling pathway,MAPK signaling pathway,HIF-1 signaling pathway,and tumor necrosis factor(TNF)signaling pathway.Animal experiments showed that Dan'e Fukang Decoction effectively inhibited the growth of ectopic tissues and alleviated pain in EMS rats.Compared with the sham surgery group,the plasma levels of interleukin(IL)-6,TNF-α,and vascular endothelial growth factor A(VEGFA)were signifi-cantly increased in the model group(P<0.01).In the ectopic tissues of the model group,the protein levels of p-STAT3,phosphorylated ex-tracellular signal-regulated kinase 1/2(p-ERK1/2),c-MYC,cyclin D1(CCND1),matrix metalloproteinase-2(MMP2),and HIF-1α were significantly upregulated(P<0.01),and the nuclear translocation of NF-κB p65 was significantly increased(P<0.05).Compared with the model group,the plasma levels of IL-6,TNF-α,and VEGFA were significantly decreased in the Dan'e Fukang Decoction group(P<0.01).The protein expression levels of p-STAT3,p-ERK1/2,c-MYC,CCND1,MMP2,and HIF-1α in the ectopic tissues were significantly downregu-lated(P<0.01),and the nuclear translocation of NF-κB p65 was significantly reduced(P<0.05).Conclusion:Dan'e Fukang Decoction can inhibit the activation of STAT3,ERK1/2,and NF-κB,regulate the expression of target proteins such as c-MYC,CCND1,MMP2,HIF-1α,and VEGFA,and inhibit the proliferation,inflammation,and angiogenesis of EMS ectopic tissues,thus exerting therapeutic effects in the treatment of EMS.关键词
