Abstract
Objective:To investigate the effects of Pingna(平纳)honey paste on the inflammation and the Toll-like receptor 4/myeloid differentia-tion factor 88/nuclear factor κB(TLR4/MYD88/NF-κB)signaling pathway in the esophageal tissue of the rat model of reflux esophagitis(RE).Methods:A rat model of reflux esophagitis was established by the method of tail-clamping stimulation+catheter balloon dilatation.The successfully modeled rats were randomized into the model,Pingna honey paste(2.9,5.8,and 11.6 g/kg),esomeprazole(4.2 mg/kg),and Xiangsha Yangwei(香砂养胃)Pills(0.4 g/kg)groups,while normal rats were selected as the control group.Rats were administrated with corresponding agents for four weeks consecutively.During the administration,the changes in body weight were observed,and the gastric emptying rate and small intestinal propulsion rate were measured.The levels of va-soactive intestinal peptide(VIP),5-hydroxytryptamine(5-HT),motilin(MTL),and gastrin(GAS)in the serum,as well as the levels of interleukin-2(IL-2)and tumor necrosis factor-α(TNF-α)in the esophageal tissue were measured by enzyme-linked immunosorbent assay.The protein levels of TLR4,MyD88,and NF-κB were determined by Western blotting.The his-topathological changes in the esophageal tissue were observed by hematoxylin-eosin staining.Glacial acetic acid was used to in-crease the capillary permeability in the abdominal cavity of mice(the OD value of peritoneal washing fluid was determined)and cotton balls were used to induce granulomas of mice(the weight of the pre-embedded cotton ball was determined).The effects of Pingna honey paste on non-specific acute inflammation and chronic inflammation of mice were observed.Results:Compared with the control group,the model group showed decreases in the gastric emptying rate and small intestinal propulsion rate(P<0.01),eleva-tions in the levels of IL-2,TNF-α,VIP,and 5-HT in the esophageal tissue(P<0.01),declines in the serum levels of GAS and MTL(P<0.01),and up-regulated protein levels of TLR4,MyD88,and NF-κB p65(P<0.01).In addition,the modeling led to inflammatory cell infil-tration in the gastric mucosal layer,basal hyperplasia of esophageal squamous epithelium,and extension of lamina propria papillae in rats.The modeling in mice increased in the OD value of peritoneal washing fluid and formation of granulomas(P<0.01).Compared with the model group,Pingna honey paste groups showed increases in the gastric emptying rate and small intestinal propulsion rate(P<0.01 or P<0.05),de-clines in the levels of IL-2,TNF-α,VIP,and 5-HT in the esophageal tissue,rises in the serum levels of GAS and MTL(P<0.01 or P<0.05),and down-regulated protein levels of TLR4,MyD88,and NF-κB p65(P<0.01 or P<0.05).Furthermore,Pingna honey paste reduced in-flammatory cell infiltration in the gastric mucosal layer,basal hyperplasia of esophageal squamous epithelium,and extension of lamina propria papillae in rats,as well as the OD value of peritoneal washing fluid and cotton ball-induced formation of granulomas in mice(P<0.01).Conclusion:Pingna honey paste can reduce inflammatory lesions of esophageal mucosa,lower the levels of inflammatory factors in the esopha-geal tissue,regulate the levels of gastrointestinal hormones,alleviate the esophageal mucosa damage caused by gastric reflux,and inhibit the activation of the TLR4/MyD88/NF-κB signaling pathway in the esophageal tissue,thus playing a role in treating RE.关键词
