科罗索酸抑制NLRP3介导的细胞焦亡减轻阿霉素诱导的心肌细胞损伤
Corosolic Acid Attenuates Doxorubicin-Induced Injury of Cardiomyocytes by Inhibiting NLRP3-Mediated Pyroptosis

Abstract
Objective:To investigate the effect and mechanism of corosolic acid(CRA)in attenuating doxorubicin(Dox)-induced injury in H9c2 cardiomyocytes.Methods:H9c2 cardiomyocytes were exposed to 1 μmol/L Dox for the modeling of cell injury and simultaneously treated with different concentrations of CRA for 24 h.The methyl thiazolyl tetrazolium assay and scanning electron microscopy were employed to examine the viability and morphology of H9c2 cardiomyocytes,respectively.The lactate dehydrogenase(LDH)release level of H9c2 cells was meas-ured,and the production of reactive oxygen species(ROS)in the cells was detected by the fluorescence probe 2',7'-dichlorofluorescein diace-tate(DCFH-DA).Western blotting was adopted to determine the protein levels of NOD-like receptor family pyrin domain-containing 3(NL-RP3),apoptosis-associated speck-like protein containing a CARD(ASC),cysteinyl aspartate-specific protease-1(CASPASE-1),interleukin(IL)-1β,and IL-18.Furthermore,the NLRP3 agonist nigericin was used to validate the relationship between CRA and NLRP3-mediated py-roptosis.Results:Compared with the normal control group,the model group showed decreased viability(P<0.01),increased LDH release(P<0.01)and ROS production(P<0.01),and up-regulated expression of NLRP3,ASC,CASPASE-1,IL-1β,and IL-18(P<0.01)in H9c2 cardiomyocytes.Compared with the model group,CRA treatment increased the viability(P<0.01),decreased the LDH release(P<0.01)and ROS production(P<0.01),and down-regulated the expression of NLRP3,ASC,CASPASE-1,IL-1β,and IL-18(P<0.01)in H9c2 car-diomyocytes.And.Conclusion:CRA can inhibit the Dox-induced injury of H9c2 cardiomyocytes by reducing ROS production and inhibiting NLRP3-mediated pyroptosis.关键词
